A study of the role of myeloid-derived growth factor on acute lung injury in vitro and In vivo

Objectives: Bone marrow-derived mesenchymal stem cells (BMSCs) are considered to have potential clinical application value in the treatment of acute lung injury (ALI). Myeloid-derived growth factor (MYDGF) can promote proliferation cell. We hypothesized that MYDGF may play a role reducing vitro and vivo through bone marrow cells. Methods: An model lipopolysaccharide (LPS)(MLE-12) was established, which divided into five groups: A: MLE-12; B: MLE-12+LPS; C: MLE-12+LPS + BMSCs; D: MYDGF; E: BMSCs MYDGF. A Cell Counting Kit-8 used detect OD value. And an ALI constructed by inducing mice with lipopolysaccharide. Forty male Balb/c were randomly control group; LPS +BMSCs +MYDGF. Specimens collected after 24 h. Hematoxylin-eosin (HE)-staining performed on tissue sections. The protein concentration alveolar lavage fluid measure bicinchoninic acid (BCA). NF-κB, p-Akt, Bax, Bcl-2 expression detected Western blotting, Enzyme linked immunosorbent assay (ELISA) serum interleukin-6, interleukin-10, TNF-α. Results: Compared group, alleviate induced ( p < .05). Conclusion: found combined effect better than using or alone. speculate pretreatment improve survival transplanted MSCs, thereby improving curative cell transplantation.
